چکیده

The tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA) caused a marked stimulation of inorganic [32P]orthophosphate incorporation into HeLa-cell phosphatidylcholine (PC), phosphatidylethanolamine (PE), and lysophosphatidylethanolamine. The increased incorporation of inorganic [32P]orthophosphate into PE and lysophosphatidylethanolamine in the presence of TPA was not associated with an increase in PE synthesis as detected by the incorporation of [3H]serine or [3H]ethanolamine. The PC-specific exchange protein from beef liver was used to insert PC labeled with [3H]choline, inorganic [32P]orthophosphate, or [14C]arachidonic acid plus [3H]palmitic acid into the outer monolayer of intact HeLa cell membranes. Radioactivity from the latter two compounds was rapidly incorporated into PE and lysophosphatidylethanolamine; the incorporation was stim ulated by TPA. It was concluded that TPA stimulated the for mation of PE by base exchange between ethanolamine and PC.

Alterations in Lipid Metabolism Induced by 12-O-Tetradecanoylphorbol- 13-acetate in Differentiating Human Myeloid Leukemia Cells1

Several aspects of lipid metabolism were evaluated ¡ndiffer entiating human myeloid leukemia (HL-60) cells after treatment with the tumor promoter 12-O-tetradecanoylphorbol-13-acetate. Modifications accompanying the phorbol ester-induced differentiation include an increase ¡nthe incorporation of ace tate into free fatty acids and neutral lipids, an increase in the amount of neutral glycerolip...

The effects of specific expression of apoptin under the control of PSES and PSA promoter on cell death and apoptosis of LNCaP cells

Objective(s): Apoptotic effect of apoptin has been demonstrated in numerous studies. However, its tumor specificity has been questioned by some reports. The aim of this study was to confine the expression of apoptin in the prostate tumor cells by inducing its gene expression under the control of a chimeric enhancer composing of prostate-specific membrane antigen (PSMA) and prostate-specific ant...

Promoter Methylation of Four Tumor Suppressor Genes in Human Papillary Thyroid Carcinoma

Background & Objective: Papillary thyroid cancer (PTC) is considered to be the most common type of thyroid malignancies. Epigenetic alteration, in which the chromatin conformation and gene expression change without changing the sequence of DNA, can occur in some tumor suppressor genes and oncogenes. Methylation is the most common type of epigenetic alterations that can be an ex...

Cytotoxic evaluation of iranian conifer extracts

Isolation and identification of some potent anti-tumor compounds from medicinal plants, has motivated researchers to screen different parts of plant species for anti-tumor effects. It has been reported that several conifer posses cytotoxic activities on some human tumor cell lines. In this study, male and female branchlets or fruit of four different species of Iranian conifers (Juniperus Sabina...

The effect of poliomyelitis virus type I (Mahoney strain) on the phospholipid metabolism of the HeLa cell.

There have been many investigations on the uptake of radioactive phosphorus by viral infected cells (1, 2), but the number of studies in which cells of animal origin are employed are few. To our knowledge, there are no studies on the effects of poliomyelitis virus on phosphorus metabolism in the HeLa cell. However, this cell has been successfully infected by the poliomyelitis virus (3, 4). In t...
